چکیده

When intact pea (Pisum sativum L.) plants are illuminated, the glycolytic enzyme phosphofructokinase is inactivated. In crude extracts the enzyme is inhibited by dithiothreitol. It would seem that this cytoplasmic enzyme, like glucose-6-P dehydrogenase, is light-inactivated when the enzymes of photosynthetic carbon metabolism are light-activated.

Light modulation of phosphofructokinase in pea leaf chloroplasts.

Chloroplastic phosphofructokinase, phosphorylase, phosphoglucomutase, and phosphoglucoisomerase in peas are light inactivated. The effect of light on phosphofructokinase is mimicked by dithiothreitol. DCMU, arsenite, and sulfite inhibit light modulation of the enzyme. No effect of light inactivation on the K(m) (fructose-6-P) or pH optima of phosphofructokinase was observed.

Inhibition by Catalase of Dark-mediated Glucose-6-Phosphate Dehydrogenase Activation in Pea Chloroplasts.

Dark activation of light-inactivated glucose-6-phosphate dehydrogenase was inhibited by catalase in a broken pea chloroplast system. Partially purified glucose-6-phosphate dehydrogenase from pea leaf chloroplasts can be inactivated in vitro by dithiothreitol and thioredoxin and reactivated by H(2)O(2). The in vitro activation by H(2)O(2) was not enhanced by horseradish peroxidase, and dark acti...

Light regulation of leaf mitochondrial pyruvate dehydrogenase complex : role of photorespiratory carbon metabolism.

Light-dependent inactivation of mitochondrial pyruvate dehydrogenase complex (mtPDC) in pea (Pisum sativum L.) leaves was further characterized, and this phenomenon was extended to several monocot and dicot species. The light-dependent inactivation of mtPDC in vivo was rapidly reversed in the dark, even after prolonged illumination. The mtPDC can be efficiently cycled through the inactivated-re...

Orthophosphate control of glucose-6-phosphate dehydrogenase light modulation in relation to the induction phase of chloroplast photosynthesis.

High concentrations of orthophosphate (Pi) inhibited CO(2)-dependent O(2) evolution and prevented the inactivation of glucose-6-P dehydrogenase by light in intact spinach and barley chloroplasts. Addition of glycerate-3-P to chloroplasts inhibited by Pi in the light, induced O(2) evolution and caused rapid inactivation of glucose-6-P dehydrogenase. The activity of phosphofructokinase detected i...

Role of nitrogen content of pea (Pisum sativum L.) on pea aphid (Acyrthosiphon pisum Harris) establishment

The leaf nitrogen content is generally accepted as an indicator of food quality and as a factor affecting host selection by phytophagous insects. The alate pea aphids (Acyrthosiphon pisum Harris, Aphididae) were given a choice among non-nodulated pea plants (Pisum sativum L.) supplied with one of four nitrate-N levels (0, 3, 15 and 30 mM). When whole plants were exposed to aphids for 7 days, th...
